    Temporal and spatial patterns of airborne pollen dispersal in six salt marsh halophytes

    Pollination is a key process in the life cycle of most plant species, but very little is known about airborne pollen dispersal in salt marsh anemophilous halophytes. We analyzed the temporal and spatial dynamics of airborne pollen dispersal, using portable volumetric pollen traps, for six anemophilous Chenopodiaceae halophytic species in tidal salt marshes. In addition, we studied the effects of air temperature and wind speed on airborne pollen dispersal. Our study shows high levels of temporal and spatial constraint on airborne pollen dispersal. We recorded the lowest airborne pollen concentrations at air temperatures lower than 22 °C and wind speeds higher than 6 m s−1. These environmental conditions set up a narrow temporal pollination window during morning hours. We also recorded severe spatial limitation for pollen dispersal in three of the six halophytes studied, which presented airborne pollen concentrations that were 9 to 53 times smaller only 5 m away from source populations than within those populations. These results show that even related species colonizing the same ecosystem may show contrasted airborne pollen spatial dispersal dynamics that depend on several environmental factors. This should be taken into account for estimates of vegetation composition based on pollen production in aerobiological and paleopalinological studies. The restricted airborne pollen dispersal recorded would promote geitonogamy and limit interpopulation genetic exchange to the dispersal of their hydrochorous fruits.The authors thank the Directorate of the Natural Parks for supporting field work conducted in this study.     Effects of heavy metal pollution on germination and early seedling growth in native and invasive Spartina cordgrasses

    Seed germination and seedling establishment are the stages most sensitive to abiotic stress in the plant life cycle. We analyzed the effects of copper, zinc and nickel on seed germination and early seedling growth of native Spartina maritima and invasive S. densiflora from polluted and non-polluted estuaries. Germination percentages for either species were not affected by any metal at any tested concentration (up to 2000 ÎŒM). However, the increase in metal concentration had negative effects on S. densiflora seedlings. The primary effect was on radicle development, representing initial seedling emergence. Spartina densiflora seedlings emerging from seeds from Tinto Estuary, characterized by high bioavailable metal loads, showed higher tolerance to metals than those from less polluted Odiel and Piedras Estuaries. Comparing our results to metal concentrations in the field, we expect S. densiflora seedling development would be negatively impacted in the most metal-polluted areas in Odiel and Tinto Estuaries.We thank the management and staff of the Odiel Marshes Natural Park for their collaboration.     Seed Viability, Spikelet Dispersal, Seed Banks and Seed Storage Requirements for Native and Invasive Cordgrasses (Genus Spartina) in Southwest Iberian Peninsula

    Seed banks play an important role in determining the spatial and temporal distribution of halophytes in salt marshes. We tested the ability of native Spartina maritima and invasive S. densiflora spikelets to disperse by flotation on water with different salinity concentrations, and the longevity of spikelet viability relative to environmental conditions including dry or wet storage at low or moderate temperatures, and aqueous salinity concentrations from fresh to sea water. We quantified pre-dispersal seed production and pre- and post-dispersal focal Spartina spp. presence in seed banks along intertidal gradients within salt marshes at the Gulf of CĂĄdiz (Southwest Iberian Peninsula). Spartina densiflora spikelets, especially from middle and high elevation marshes, showed greater ability than S. maritima to remain afloat, which suggests this species could be dispersed over longer distances. Wet-stored Spartina seeds were able to maintain viability for months, while seeds under dry storage rapidly lost viability. This decline was most significant for S. maritima, and for seeds stored at moderate temperatures. Storage of spikelets under wet and cold conditions optimized spikelet viability. Native S. maritima did not establish transient or persistent seed banks, while invasive S. densiflora established transient seed banks mainly at higher marsh elevations. Our results on the dynamics of seed dispersal and seed banks and seedling recruitment provide fundamental knowledge that can be applied for conservation of native S. maritima, management of invasive S. densiflora, and ecological restoration of tidal salt marshes.Funding for open access publishing: Universidad de Sevilla / CBUA Open Access funding enabled and organized by Universidad de Sevilla.     Accuracy of Hidden Markov Models in Identifying Alterations in Movement Patterns during Biceps-Curl Weight-Lifting Exercise

    This paper presents a comparison of mathematical and cinematic motion analysis regarding the accuracy of the detection of alterations in the patterns of positional sequence during biceps-curl lifting exercise. Two different methods, one with and one without metric data from the environment, were used to identify the changes. Ten volunteers performed a standing biceps-curl exercise with additional loads. A smartphone recorded their movements in the sagittal plane, providing information on joints and barbell sequential position changes during each lift attempt. An analysis of variance revealed significant differences in joint position (p < 0.05) among executions with three different loads. Hidden Markov models were trained with data from the bi-dimensional coordinates of the joint positional sequence to identify meaningful alteration with load increment. Tests of agreement tests between the results provided by the models with the environmental measurements, as well as those from image coordinates, were performed. The results demonstrated that it is possible to efficiently detect changes in the patterns of positional sequence with and without the necessity of measurement and/or environmental control, reaching an agreement of 86% between each other, and 100% and 86% for each respective method to the results of ANOVA. The method developed in this study illustrates the viability of smartphone camera use for identifying positional adjustments due to the inability to control limbs in an adequate range of motion with increasing load during a lifting task.info:eu-repo/semantics/publishedVersio

    Statistical validation of Aeolus L2A particle backscatter coefficient retrievals over ACTRIS/EARLINET stations on the Iberian Peninsula

    The Global Observing System (GOS) has encountered some limitations due to a lack of worldwide real-time wind measurements. In this context, the European Space Agency (ESA) has developed the Aeolus satellite mission, based on the ALADIN (Atmospheric Laser Doppler Instrument) Doppler wind lidar; this mission aims to obtain near-real-time wind retrievals at the global scale. As spin-off products, the instrument retrieves aerosol optical properties such as particle backscatter and extinction coefficients. In this work, a validation of Aeolus reprocessed (baseline 10) co-polar backscatter coefficients ( part Aeolus) is presented through an intercomparison with analogous ground-based measurements taken at the ACTRIS (Aerosols, Clouds and Trace gases Research InfraStructure Network)/EARLINET (European Aerosol Research Lidar Network) stations of Granada (Spain), Évora (Portugal) and Barcelona (Spain) over the period from July 2019 until October 2020. Case studies are first presented, followed by a statistical analysis. The stations are located in a hot spot between Africa and the rest of Europe, which guarantees a variety of aerosol types, from mineral dust layers to continental/anthropogenic aerosol, and allows us to test Aeolus performance under different scenarios. The so called Aeolus-like profiles ( part Aeolus like;355) are obtained from total particle backscatter coefficient and linear particle depolarization ratio ( part linear) profiles at 355 and 532 nm measured from the surface, through a thorough bibliographic review of dual-polarization measurements for relevant aerosol types. Finally, the study proposes a relation for the spectral conversion of part linear, which is implemented in the Aeolus-like profile calculation. The statistical results show the ability of the satellite to detect and characterize significant aerosol layers under cloud-free conditions, along with the surface effect on the lowermost measurements, which causes the satellite to largely overestimate copolar backscatter coefficients. Finally, the Aeolus standard correct algorithm middle bin (SCAmb) shows a better agreement with ground-based measurements than the standard correct algorithm (SCA), which tends to retrieve negative and meaningless coefficients in the clear troposphere. The implementation of Aeolus quality flags entails a vast reduction in the number of measurements available for comparison, which affects the statistical significance of the results

    Cytokine and antibody responses to Plasmodium falciparum in naĂŻve individuals during a first malaria episode: effect of age and malaria exposure.

    Age- and exposure-dependent immune responses during a malaria episode may be key to understanding the role of these factors in the acquisition of immunity to malaria. Plasma/serum samples collected from naĂŻve Mozambican children (n = 48), European adults (naĂŻve travelers, n = 22; expatriates with few prior malaria exposures, n = 15) and Mozambican adults with long-life malaria exposure (n = 99) during and after a malaria episode were analyzed for IgG against merozoite proteins by Luminex and against infected erythrocytes by flow cytometry. Cytokines and chemokines were analyzed in plasmas/sera by suspension array technology. No differences were detected between children and adults with a primary infection, with the exception of higher IgG levels against 3D7 MSP-142 (P = 0.030) and a P. falciparum isolate (P = 0.002), as well as higher IL-12 (P = 0.020) in children compared to other groups. Compared to malaria-exposed adults, children, travelers and expatriates had higher concentrations of IFN-Îł (P≀0.0090), IL-2 (P≀0.0379) and IL-8 (P≀0.0233). Children also had higher IL-12 (P = 0.0001), IL-4 (P = 0.003), IL-1ÎČ (P = 0.024) and TNF (P = 0.006) levels compared to malaria-exposed adults. Although IL-12 was elevated in children, overall the data do not support a role of age in immune responses to a first malaria episode.     High antibody responses against Plasmodium falciparum in immigrants after extended periods of interrupted exposure to malaria.

    Background Malaria immunity is commonly believed to wane in the absence of Plasmodium falciparum exposure, based on limited epidemiological data and short-lived antibody responses in some longitudinal studies in endemic areas. Methods A cross-sectional study was conducted among sub-Saharan African adults residing in Spain for 1 up to 38 years (immigrants) with clinical malaria (n=55) or without malaria (n=37), naĂŻve adults (travelers) with a first clinical malaria episode (n=20) and life-long malaria exposed adults from Mozambique (semi-immune adults) without malaria (n=27) or with clinical malaria (n=50). Blood samples were collected and IgG levels against the erythrocytic antigens AMA-1 and MSP-142 (3D7 and FVO strains), EBA-175 and DBL-α were determined by Luminex. IgG levels against antigens on the surface of infected erythrocytes (IEs) were measured by flow cytometry. Results Immigrants without malaria had lower IgG levels than healthy semi-immune adults regardless of the antigen tested (P≀0.026), but no correlation was found between IgG levels and time since migration. Upon reinfection, immigrants with malaria had higher levels of IgG against all antigens than immigrants without malaria. However, the magnitude of the response compared to semi-immune adults with malaria depended on the antigen tested. Thus, immigrants had higher IgG levels against AMA-1 and MSP-142 (P≀0.015), similar levels against EBA-175 and DBL-α, and lower levels against IEs (P≀0.016). Immigrants had higher IgG levels against all antigens tested compared to travelers (P≀0.001), both with malaria. Conclusions Upon cessation of malaria exposure, IgG responses to malaria-specific antigens were maintained to a large extent, although the conservation and the magnitude of the recall response depended on the nature of the antigen. Studies on immigrant populations can shed light on the factors that determine the duration of malaria specific antibody responses and its effect on protection, with important implications for future vaccine design and public health control measures

    Immune and spermatogenesis-related loci are involved in the development of extreme patterns of male infertility

    We conducted a genome-wide association study in a large population of infertile men due to unexplained spermatogenic failure (SPGF). More than seven million genetic variants were analysed in 1,274 SPGF cases and 1,951 unaffected controls from two independent European cohorts. Two genomic regions were associated with the most severe histological pattern of SPGF, defined by Sertoli cell-only (SCO) phenotype, namely the MHC class II gene HLA-DRB1 (rs1136759, P = 1.32E-08, OR = 1.80) and an upstream locus of VRK1 (rs115054029, P = 4.24E-08, OR = 3.14), which encodes a protein kinase involved in the regulation of spermatogenesis. The SCO-associated rs1136759 allele (G) determines a serine in the position 13 of the HLA-DR beta 1 molecule located in the antigen-binding pocket. Overall, our data support the notion of unexplained SPGF as a complex trait influenced by common variation in the genome, with the SCO phenotype likely representing an immune-mediated condition.     A clinically compatible drug-screening platform based on organotypic cultures identifies vulnerabilities to prevent and treat brain metastasis

    We report a medium‐throughput drug‐screening platform (METPlatform) based on organotypic cultures that allows to evaluate inhibitors against metastases growing in situ. By applying this approach to the unmet clinical need of brain metastasis, we identified several vulnerabilities. Among them, a blood–brain barrier permeable HSP90 inhibitor showed high potency against mouse and human brain metastases at clinically relevant stages of the disease, including a novel model of local relapse after neurosurgery. Furthermore, in situ proteomic analysis applied to metastases treated with the chaperone inhibitor uncovered a novel molecular program in brain metastasis, which includes biomarkers of poor prognosis and actionable mechanisms of resistance. Our work validates METPlatform as a potent resource for metastasis research integrating drug‐screening and unbiased omic approaches that is compatible with human samples. Thus, this clinically relevant strategy is aimed to personalize the management of metastatic disease in the brain and elsewhere
